بررسی کارایی جدایه های Trichoderma spp. در کنترل بیماری پوسیدگی زغالی سویا ناشی از Macrophomina phaseolina در شرایط گلخانه
بیماری پوسیدگی زغالی ناشی از Macrophomina phaseolina، از معمولترین بیماریهای سویا در بسیاری از نقاط دنیا میباشد.بهدلیل خاکزی بودن قارچ بیمارگر و توان بالای ساپروفیتی آن در خاک، عدم کنترل مؤثر بیماری و ایجاد آلودگیهای زیستمحیطی در اثر استفاده از سموم شیمیایی، از عوامل آنتاگونیست طبیعی مانند جدایههای قارچی متعلق به جنس Trichoderma برای مهارزیستی این بیماری میتوان استفاده کرد. در این پژوهشتأثیر دو جدایه متعلق بهگونههای Trichoderma harzianum و Trichoderma atroviride در کنترل M. phaseolina و همچنین القای مقاومت گیاه نسبت به این بیمارگر در شرایط گلخانهای مورد بررسی قرار گرفت. نتایج نشان داد که در تیمارهای آلوده سازی خاک با بیمارگر، جدایهی T. harzianum در خاکهای سترون و غیرسترون بهترتیب موجب کاهش بیماری بهمیزان 62 و 65 درصد شد. جدایه T. atroviride نیز بیماری را در خاکهای سترون و غیرسترون، بهترتیب 59 و 62 درصد کاهش داد. تلفیق دو جدایهی تریکودرما با هم، بهترتیب بیماری را 67 و 70 درصد را کاهش دادند.براساس نتایج بهدست آمده، در تیمارهای آلودهسازی نوک ساقهی سویا با بیمارگر، گونهی T. harzianum در خاکهای سترون و غیرسترون بهترتیب موجب 56 و 58 درصد کاهش بیماری شد. جدایهی T. atroviride نیز در خاکهای سترون و غیرسترون بهترتیب موجب کاهش بیماری بهمیزان 52 و 53 درصد شد. تلفیق دو جدایهی تریکودرما با هم، بهترتیب بیماری را 60 و 63 درصد کاهش دادند.در مجموع، در این پژوهش، جدایههای تریکودرما در خاک غیرسترون کارایی بیشتری در کاهش بیماری و القای مقاومت نشان دادند. جدایههای تریکودرما در تیمارهای آلودهسازی خاک با بیمارگر تأثیر بیشتری در کاهش بیماری داشتند. جدایه T. harzianum نسبت به جدایه T. atroviride موجب کاهش بیشتر بیماری در هر دو روش کاربرد بیمارگر شده است. تلفیق دو جدایه تریکودرما نیز تأثیر بیشتری در کاهش بیماری و القای مقاومت داشتند.

کنترل بیولوژیک بیماری لکه قهوهای برنج ناشی از Bipolaris victoriae با استفاده از جدایههای قارچی در شرایط آزمایشگاه و گلخانه
بیماری لکه قهوهای برنج که توسط گونههای مختلفی از Bipolaris ایجاد میشود یکی از مهمترین بیماریهای برنج در ایران و جهان است. در این تحقیق، از مجموع ۲۲۰ نمونه آلوده جمعآوری شده از مزارع برنج استان گیلان، با استفاده از محیطهای کشت سیبزمینی، دکستروز آگار و آب آگار، ۸۰ جدایه قارچی جداسازی شد. برای شناسایی این قارچها، خصوصیات مورفولوژیکی آنها مورد استفاده قرار گرفت و قارچهای شناسایی شده به گونههای Bipolaris victoriae(۲۴ جدایه)، Bipolaris oryzae(۱۲ جدایه)، Alternaria tenuissima(۱۱ جدایه)، Preussia sp. (۸ جدایه)، Fusarium verticillioides(۶ جدایه)، Alternaria infectoria (۷ جدایه)، Alternaria citri(۹ جدایه)، Trichoderma harzianum(یک جدایه) و Trichoderma virens(۲ جدایه) تعلق داشتند. بیماریزایی تمامی جدایههای Bipolaris victoriae و Bipolaris oryzae به اثبات رسید و از بین جدایههای قارچی متعلق به جنسهای قارچی دیگر20 جدایه که در برنج ایجاد بیماری نکرده یا قدرت بیماریزایی آنها بسیار کم بود برای بررسیهای مهار زیستی انتخاب شدند. در آزمایشگاه از روشهای مختلفی مانند مطالعه بازدارندگی رشد victoriaeB. با استفاده از عصارهی کشت، کشت متقابل قارچهای آنتاگونیست مورد نظر و victoriaeB. بهروش کشت اسلاید و اثر متابولیتهای فرار در بازدارندگی رشد این قارچ استفاده شد. T. harzianum بهعنوان مؤثرترین جدایه در مهار رشد میسلیومی عامل بیماری در آزمایشگاه شناخته شد. در گلخانه همهی جدایهها به استثنای A. tenuissima و F. verticillioides شدت بیماری لکه قهوهای را کاهش دادند که در این بین، جدایهی Preussia sp. با کاهش شدت بیماری به میزان 4/33% مؤثرترین آنتاگونیست در بررسیهای گلخانهای تشخیص داده شد و در رتبههای بعدی بهترتیب جدایههای T. harzianum، A. citri، A. infectoria و T. virensاز بیشترین کارایی در کنترل بیماری برخوردار بودند. تجزیهی واریانس و مقایسهی میانگین صفات بهروش حداقل اختلاف معنیدار (LSD)، تفاوت معنیدار قارچهای مورد مطالعه را از نظر کارایی در کنترل بیماری لکه قهوهای برنج اثبات کرد.

مطالعهی اثر بیوکنترلی سویههای سودوموناس فلورسنت مولد 2 و 4- دی استیل فلوروگلوسینول بر نماتد ریشه گرهی در ریزوسفر ارقام گوجهفرنگی
نماتد مولد ریشه گرهی Meloidogyne javanica یکی از عوامل مهم بیمارگر گوجهفرنگی در کشور بهشمار میرود. در این تحقیق کنترل بیولوژیک این نماتد توسط سویههای UTPF68 و UTPF101باکتری Pseudomonas fluorescens، مورد ارزیابی قرار گرفت. ابتدا سویهها از نظر دارا بودن ژن phlD از طریق واکنش زنجیرهای پلیمراز (PCR) مورد بررسی قرار گرفتند. سپس توانایی تولید متابولیتهای ضد میکروبی توسط این باکتریها و تأثیر آنها بر تفریخ تخم و فعالیت لارو سن دوم نماتد در شرایط آزمایشگاه و اثر بیوکنترلی آنها بر فعالیت نماتد در ریزوسفر چهار رقم گوجهفرنگی در شرایط گلخانه براساس طرح بلوکهای کامل تصادفی با چهار تکرار مورد بررسی قرار گرفت. نتایج واکنش زنجیرهای پلیمراز نشان داد که هر دو سویه واجد ژن phlD بوده و قطعه DNA با طول 629 جفت باز در آنها تکثیر شد. سویههای باکتریایی قادر به تولید آنتی بیوتیک DAPG، سیانید هیدروژن و پروتئاز در شرایط آزمایشگاهی بودند. سویه UTPF68 نسبت به سویه UTPF101 توانایی بیشتری در عدم تفریخ تخم نماتد داشت، درحالیکه میزان مرگ و میر لارو سن دوم در هر دو سویه یکسان بوده است. نتایج گلخانهای نشان داد که سویههای باکتری موجب افزایش شاخصهای رشدی ارقام و کاهش فعالیت نماتد در ریزوسفر آنها شدند. میزان افزایش رشد یا کنترل نماتد بسته به رقم گوجهفرنگی و سویه باکتری متفاوت بود.

کارایی تثبیتکنندههای شیمیایی قارچ آنتاگونیست Talaromyces flavus در کنترل بیولوژیک بیماری مرگ گیاهچه رایزوکتونیایی گوجهفرنگی و خیار گلخانهای
قارچ آنتاگونیست Talaromyces flavus از مهمترین عوامل کنترل بیولوژیک بیماریهای گیاهی خاکزاد مانند مرگ گیاهچه محسوب میشود. در این پژوهش، برای افزایش کارایی زادمایه جدایههای T. flavusبهدست آمده از گلخانههای خیار و مزارع گوجهفرنگی، پنج تثبیتکننده شیمیایی این قارچ بررسی شدند. براساس نتایج تحقیقات پیشین، مؤثرترین بسترها از لحاظ رشد، اسپورزایی و پایداری برای جدایههای T. flavus مربوط به خیار گلخانهای و گوجهفرنگی، ترکیب سبوس برنج و خاک پیت بود. برای هر گیاه، آزمایشی در قالب طرح آماری بلوکهای کامل تصادفی با هفت تیمار و سه تکرار انجام شد. تیمارهای آزمایشی شامل پنج تثبیت کننده مختلف (آمینوفنل، دیسیکلوسرین، سولفات منیزیم، کربوکسیمتیلسلولز و نیترات سدیم)، شاهد آلوده و شاهد سالم بودند. ارزیابی تیمارهای مختلف، براساس تعداد گیاهچههای سالم انجام گرفت. نتایج کلی این پژوهش نشان داد که زادمایههای T. flavus در برگیرنده تثبیتکنندههای نیترات سدیم، دیسیکلوسرین و کربوکسیمتیلسلولز در مقایسه با آمینوفنل و سولفات منیزیم کارایی بیشتری در کنترل بیماری مرگ گیاهچه داشتند.

خاصیت ضدباکتریایی گیاه صبر زرد علیه باکتری Pectobacterium carotovorum subsp. carotovorum عامل پوسیدگی نرم سیبزمینی
در این مطالعه اثر بازدارندگی عصارهی اتانولی، متانولی، استونی و کلروفرمی بخشهای مختلف برگ گیاه صبر زرد شامل بخش پارانشیمی، ژل و شیرابه بر باکتری عامل بیماری پوسیدگی نرم غده سیبزمینی Pectobacterium carotovorum subsp. carotovorum بهروش نشر در آگار بررسی شد. همچنین اثر باکتریایستایی و باکتریکشی عصارههای مذکور در شرایط آزمایشگاهی مورد ارزیابی قرار گرفت. تمام بخشهای برگ گیاه صبر زرد حاوی ترکیبات ضدباکتریایی بودند. حلال متانول، عصارههایی با بازدارندگی نسبتا بیشتری را ایجاد نمود. اثر باکتریایستایی و باکتریکشی عصارههای کلروفرمی در قیاس با عصاره سایر حلالها، ضعیفتر بود. اثر باکتری ایستایی و باکتریکشی ژل روی باکتری مورد مطالعه از سایر قسمتهای برگی بیشتر بود. در کل، 31 ترکیب از مواد گیاهی گیاه صبر زرد با روش کروماتوگرافی گازی استخراج شد که 25 مورد آن در شیرابهی گیاه یافت شد. مواد ضدباکتری چون سینامیک اسید، تترادکانوئیک اسید، 2-هیدروکسی اتیل پروپیونیت، سیتوسترول، لوپئول و کاروون و 1-هپتانول-2-پروپیل و 1و2-بنزن دیکربوکسیلیک اسید در قسمتهای مختلف برگ گیاه صبر زرد بهعنوان مواد ضدباکتریایی وجود داشتند. با توجه به توان ضدباکتریایی عصارههای برگ گیاه صبر زرد، این خاصیت را میتوان برای مهار باکتری پوسیدگی نرم باکتریایی سیبزمینی مد نظر قرار داد.

مطالعهی میزان کلونیزاسیون ریشه و کاهش بیماری زنگ برگی گندم در برهمکنشهای جدایه باکتری Pseudomonas fluorescens، رقم گندم و قارچPuccinia triticina
در این پژوهش قابلیت میزان کلونیزه شدن ریزوسفر سه رقم گندم (بولانی، روشن و فورنو) توسط دو جدایه باکتری سودمند Pseudomonas fluorescens PF153mcherry و P. fluorescens CHA0gfp2 به روش آغشتهسازی بذر در کاهش بیماری زنگ برگی ناشی از Puccinia triticina در شرایط گلخانه مورد بررسی قرار گرفت. نتایج نشاندهنده کاهش شدت بیماری روی برگها در تعاملهای خاصی از جدایه باکتری و رقم گندم بود. این کاهش بیماری بهطور ویژهای در تعامل جدایهی CHA0gfp2و رقم فورنو با 74/69% کاهش جوش روی برگها مشاهده شد. دو رقم بولانی و فورنو فاقد زادمایه باکتری، بیشترین میزان آلودگی به زنگ را داشتند. بررسی اثر متقابل جدایه باکتری، رقم گندم و بیمارگر نشان داد که در تعاملهای جدایهی PF153mcherry با رقم روشن آلوده و غیرآلوده به زنگ، بیشترین جمعیت باکتریایی روی ریشه وجود دارد (بهترتیب 2/7801 و 4/8154 یاخته باکتری در میلیگرم وزن خشک ریشه). کمترین تعداد یاخته باکتری روی ریشه در تعامل جدایه CHA0gfp2با رقم روشن آلوده و غیرآلوده به زنگ بهترتیب با جمعیتی برابر با 2/938 و 3/887 یاخته باکتری در میلیگرم وزن خشک ریشه مشاهده شد. تعامل بیمارگر با هر سه رقم گندم باعث افزایش معنیدار فعالیت فنیل آلانین آمونیالیاز در برگها شد. همچنین آلودگی به زنگ در افزایش فعالیت پراکسیداز تفاوت معنی داری نشان داد. در نهایت بین میزان کلونیزاسیون باکتری (تعداد یاخته باکتری در میلیگرم وزن خشک ریشه) با شدت بیماری زنگ برگی همبستگی منفی مشاهده شد. همچنین همبستگی مثبت بین آنزیم فنیل آلانین آمونیالیاز و شدت بیماری وجود داشت. نتایج این تحقیق نشاندهنده برقراری تعامل سودمند بین جدایه ریزوباکتری-رقم گندم است که در راستای القای مقاومت و مهار بیمارگرهای برگی در سیستم کشاورزی پایدار امیدوارکننده بود.

اثر چند ترکیب گیاهی در مهارآلودگیهای قارچی و باکتریایی در محیط کشتهای ریزازدیادی گیاهان
در کشت درونشیشهای گیاهان پیازی، وجود آلودگیهای قارچی و باکتریایی از مشکلات اساسی میباشند که میتوانند کارایی این نوع روشهای تکثیری را تحت تأثیر قرار دهند. بهمنظور بررسی تاثیر غلظتهای مختلف ترکیبات گیاهی در رفع این قبیل آلودگیها از محیطکشت گیاه پیازی لاله واژگون، سه آزمایش بهطور جداگانه در قالب طرح کاملا تصادفی در پنج تکرار انجام شد. تیمارهای آزمایشی شامل غلظتهای مختلف اس- کاروون، تیمول و کارواکرول بود. بین تیمارها از لحاظ بازدارندگی از آلودگیهای قارچی و باکتریایی، اختلاف معنیداری در سطح احتمال پنج درصد وجود داشت، بهطوریکه غلظت ppm100 کارواکرول و ppm150 تیمول بازدارندگی کامل از رشد قارچ Alternaria sp. را نشان داد. همچنین رشد باکتری Pectobacteriumcarotovorum subsp. carotovorumبا چگالی نوری 2/0-1/0 در غلظت ppm200 کارواکرول و تیمول و نیز در چگالی نوری 2 با غلظت ppm300 تیمول و ppm400 کارواکرول بهطور کامل متوقف شد. بنابراین بهنظر میرسد که استفاده از ترکیبات گیاهی تیمول و کارواکرول میتواند در کنترل آلودگیهای قارچی و باکتریایی محیط کشت ریزازدیادی گیاهان پیازی از جمله لاله واژگون مفید واقع شود.

مقایسهی نرخ شکارگری کنهی Amblyseius swirskii با تغذیه از کنههایPhyllocoptes adalius و Tetranychus urticae در شرایط آزمایشگاهی
نرخ شکارگری و ترجیح غذایی، یکی از پدیدههایی است که در بررسی کارایی و انتخاب دشمنان طبیعی، باید مورد توجه قرار گیرد. کنهی swirskiiAthias-Henriot (Acari: Phytoseiidae) Amblyseiusیکی از کنههای شکارگر خانوادهی فیتوزئید میباشد که میتواند از تعداد زیادی از بندپایان کوچک و گرده گیاهان تغذیه کند. در این تحقیق، ظرفیت شکارگری این کنه فیتوزئید در کنترل دو گونهی مهم از آفات گلخانه، کنهی تارتن دو نقطهایTetranychusurticae Koch (Acari: Tetranychidae) و کنهی گالزای اریوفیدهPhyllocoptesadaliusKeifer (Acari: Eriophyoidea) در شرایط آزمایشگاهی مورد بررسی قرار گرفت. آزمایش با کنههای بالغ شکارگر در دمای5/0± 25 درجهی سلسیوس، رطوبت نسبی 70-75 درصد و دورهی نوری 16 ساعت روشنایی و 8 ساعت تاریکی درون قفسهای مانگر انجام شد. هر کنهی بالغ روزانه با 10 عدد کنهی تارتن و 20 عدد کنهی اریوفیده تغذیه شدند. نرخ خالص شکارگری (C0) با تغذیه از کنهی تارتن و کنهی گالزا بهترتیب 183 و 420 کنه به ازای هر فرد ماده محاسبه گردید که نشاندهندهی حد بالایی از کنترل این آفت است. بیشترین میزان تغذیه در دورهی تخمریزی انجام گرفت. نتایج این پژوهش نشان میدهد کنهی A. swirskii قادر به تغذیه از هر دو کنه طعمه بوده و میتواند در کنترل زیستی این آفات نقش داشته باشد.

بررسی تاثیر محیطهای غذایی طبیعی روی میزان تولید و زندهمانی اسپور و بلاستوسپورهای قارچ Metarhizium anisopliae
در این تحقیق میزان تولید و جوانه زنی اسپور و بلاستوسپورهای قارچ Metarhizium anisopliae که روی بذرهای مختلف تولید شده بودند، مورد ارزیابی قرار گرفت. آزمایشها بهصورت فاکتوریل و در قالب طرح کاملاً تصادفی که دارای 9 تیمار در سه تکرار بودند، انجام شد. نتایج اختلاف معنیداری را بین تعداد کنیدی تولید شده، دو هفته پس از کشت نشان داد (F= 4.66, df= 7, 16, p<0.01). بیشترین میانگین تولید محصول بهمیزان 108×1/3 و 108×3 کنیدی/گرم متعلق به آرد برنج و محیط کشت SDA (شاهد) بود. کمترین میانگین تولید محصول بهمیزان 106×4 کنیدی/گرم متعلق به سبوس گندم بود. همچنین اختلاف معنیداری بین میزان تولید بلاستوسپور بعد از 4 روز کشت در محیطهای مایع مشاهده شد (F= 2.57, df=8, 18, p<0.05). بیشترین و کمترین میزان تولید بلاستوسپور بهمیزان 108×1/6 و 108×5/4 در هر میلیلیتر بهترتیب متعلق به عصارهی سبوس گندم+سبوس برنج+مخمر و سبوس برنج+مخمر بودند. تجزیهی واریانس برای تیمارهای مواد افزودنی شیمیایی و عناصر غذایی که در محیطهای کشت مایع اضافه شده بودند نشان داد که اختلاف معنیداری بین تیمارها وجود دارد (F=5.02, df= 13, 29, p<0.01) بیشترین میزان تولید بلاستوسپور در هر میلیلیتر 108×6/6، 108×1/6 و 108×7/5 بهترتیب متعلق به تیمار عصارهی سبوس برنج+سبوس گندم+مخمر+آب پنیر؛ تیمار عصارهی سبوس برنج+عصارهی سبوس گندم+مخمر و تیمار عصارهی سبوس گندم+عصارهی سبوس برنج، سولفات منیزیوم و کمترین میزان محصول تولید شده متعلق به تیمار آب انگور بود. نتایج نشان داد که میانگین درصد جوانهزنی کنیدیهای M. anisopliae بعد از یک، دو و سه ماه که در شرایط 4 و 25 درجهی سلسیوس نگهداری شده بودند، تفاوت معنیداری با هم داشتند. درصد جوانهزنی در تمام تیمارهایی که در یخچال نگهداری شده بودند بیشتر از تیمارهایی بودند که در دمای معمولی اتاق حفظ شده بودند.
