اثر آنتاگونیستی جدایه های قارچی و ترکیبات زیستی در کنترل نماتود سیستی چغندرقند Heterodera schachtii
برای بررسی کنترل بیولوژیک نماتود سیستی چغندرقند Heterodera schachtii، اثر جدایههای قارچیTrichoderma harzianum، Talaromyces flavus، Fusarium solani وPochonia chlamydosporia var. chlamydosporia جدا شده از سیستهای آلوده در استان اصفهان، و ترکیبات زیستی تجاری ماری گُلد (محصول شرکت ABPL کشور هندوستان) و تریکومیکس اچ. وی.TRICHO-MIX H.V.) بـــا مـــاده مـــؤثره T. harzianum T39 محصــول شــرکت فنآوران حیات سبز)، در قالب طرح کاملاً تصادفی با 9 تیمار و پنج تکرار در گلخانه، طی سالهای 93-1392 مورد بررسی قرار گرفت. یک نمونه از هر یک از جدایههای قارچ تکثیر یافته روی گندم سترون، بهمیزان 10 گرم و ترکیبات زیستی ماری گُلد و تریکومیکس اچ. وی. بهترتیب بهمیزان 2/0 و 2 گرم، به گلدانهای حاوی خاک مزرعه با آلودگی 67/0عدد سیست و 12 عدد تخم و لارو سن دو نماتود در یک گرم خاک افزوده و در دمای 2±25 درجۀ سلسیوس و رطوبت نسبی 10±70 درصد بهمدت 90 روز نگهداری شد. تجزیۀ واریانس با نرم افزارSAS 9.1 و میانگین صفات مورد مطالعه در مقایسه با خاک سترون، بهوسیلۀ آزمون چند دامنهای دانکن ارزیابی شد. نتایج نشان داد که از نظر جمعیت نهایی بین تیمارها در مقایسه با تیمار نماتود بهتنهایی، تفاوت معنیداری وجود دارد. بهطوریکه جدایه T. harzianum 128، ماری گُلد، T. harzianum 93، TRICHO-MIX H.V. ،F. solani، T. flavus 94،T. flavus 134 و P. chlamydosporia بهترتیب بهمیزان 08/65، 72/60، 08/53، 99/47، 81/45، 17/42، 45/41 و 35/20 درصد، جمعیت نهایی نماتود سیستی چغندرقند را کاهش دادند. بوتههای چغندرقند کشت شده در خاک سترون و مزرعه، از نظر شاخصهای رشدی طول، وزن تر و خشک ریشه و اندامهای هوایی، دارای اختلاف معنیداری بودند. قارچهای فوزاریوم و پوکونیا، نسبت به سایر تیمارها، باعث رشد بیشتر بوتههای چغندرقند شدند.

افزایش تولید سورفکتین در باکتری Bacillus subtilis UTB96 با استفاده از بهینهسازی فرایند فرمانتاسیون
در میان باکتریهای زیست مهارگر، باکتری Bacillus subtilis در تولید طیف وسیعی از ترکیبات ضدمیکروبی مطرح میباشد که از این میان، بیوسورفکتانتها از جمله سورفکتین از اهمیت ویژهای برخورد است. سورفکتانتهایی که بهوسیلۀ میکروارگانیسمها تولید میشوند بهدلیل سمیّت پایین و خواص منحصر به فرد، کاربردهای متعددی دارند اما بهدلیل هزینه بالای تولید و خالصسازی، تولید در بازده بالا هنوز میسر نشده است. در این تحقیق بهمنظور ارزیابی تولید سورفکتین در جدایه مذکور، شناسایی ژن تولید کننده آن (srf) و تعیین مقدار سورفکتین تولید شده بهترتیب از روشهای متلاشی شدن قطره (Drop collapse)، روش مولکولی واکنش زنجیرهای پلیمراز (PCR) و آنالیز کروماتوگرافی مایع با کارایی بالا (HPLC) استفاده شد. بررسی مولکولی با آغازگر اختصاصی نشان داد که این جدایه دارای ژنsrf میباشد. برای بهینهسازی شرایط عملیاتی، اثر شرایط فیزیک وشیمیایی و القا کنندههای مختلف بر میزان زیست توده و سورفکتین خام تولیدی بررسی شد. شرایط بهینه فیزیکی و شیمیایی بهدست آمده شامل: مدّت زمان نگهداری کشت 96 ساعت، دمای 30 درجۀ سلسیوس، اسیدیته 7 و دور شیکر rpm 250 میباشد. از میان القاکنندهها که شامل منابع کربن، منابع نیتروژن و اسیدهای آمینه، بیشترین مقدار تولید سورفکتین پس از تیمار محیط کشت با گلوکز بهمقدار 243/1 گرم بر لیتر محیط کشت میباشد. پس از آن تأثیر عناصر القاکنندهای که بیشترین تأثیر را در افزایش تولید سورفکتین داشتهاند، بهصورت توأم بررسی شد. نتایج نشان داد بیشترین مقدار تولید سورفکتین mg/l 055/5 در حضور یونهای آهن، منیزیم و منگنز بهصورت توأم میباشد. همچنین مقدار و نوع منابع غذایی تأثیر بهسزایی در بازده تولید سورفکتین دارد. بنابر این تغییر در مقدار و ترکیب القا کنندهها روشی اقتصادی برای تولید تجاری این ماده محسوب میشود.

ترجیح میزبانی و سوئیچینگ زنبورTrissolcus vassilievi بین دو جمعیت میزبان
انتخاب میزبان بهوسیلۀ پارازیتوییدها اثرات حیاتی روی شایستگی آنها دارد. زنبور پارازیتوییدTrissolcus vassilievi (Mayr) (Hym., Platygastridae)، یکی از گونههای با پتانسیل زیستی بالا در کنترل سن گندم میباشد. بررسیها نشان داده است که تفاوتهای جزئی بین جمعیتهای مختلف میزبان میتواند موجب برتری نسبی پارازیتوییدهایی شود که در میزبانهای بزرگتر نشوونما مینمایند. در این تحقیق، ترجیح میزبانی و رفتار سوئیچینگ دو جمعیت زنبور T. vassilievi به دو جمعیت میزبان در نسبتهای 5:1، 4:2، 3:3، 2:4 و 1:5 دسته تخم بررسی شد. جمعیتهای تبریز و ورامین هم برای میزبان و هم برای زنبور بهکار رفتند. اندازهگیری قطر و وزن تخم سن گندم نشان داد که وزن تخم بهطور معنیداری در جمعیت تبریز کمتر از جمعیت ورامین بود.همچنین زنبورهای ورامین بزرگتر از زنبورهای تبریز و زنبورهای حاصل از تلاقی آنها، نزدیک به جمعیت مادری بودند. تجزیه دادههای ترجیح میزبانی و بررسی سوئیچینگ نشان داد که با توجه به مقادیر c1 (شاخص ترجیح سن گندم تبریز) و c2 (شاخص ترجیح سن گندم ورامین)، جمعیت ورامین زنبور T. vassilievi ترجیح منفی به تخم سن تبریز و ترجیح مثبت به تخم سن ورامین دارد، اما آزمون t مشخص کرد که این ترجیح معنیدار نیست. تجزیۀ رگرسیون خطی نیز نشان داد که تغییرات c با نسبت دو میزبان در هر دو مورد معنیدار است که مؤید وجود سوئیچینگ میباشد. در مجموع میتوان گفت که زنبور تفاوتی بین تخمهای دو جمعیت قائل نشده و تفاوتهای بین جمعیتی میزبان درحدی نیست که ترجیح میزبانی پارازیتویید را تحت تأثیر قرار دهد.

تأثیر جدایه های Pseudomonas fluorescens بر شدت بیماری، صفات فیزیولوژیکی و بیوشیمیایی در غدههای بذری تولیدی ارقام سیبزمینی آلوده به Rhizoctonia solani در شرایط گلخانه
بهمنظور ارزیابی تأثیر تیمار جدایههای باکتری Pseudomonas fluorescens بر ویژگیهای فیزیولوژیکی، بیوشیمیایی و شدت بیماری غدههای بذری سیبزمینی در حضور عامل بیماریزا Rhizoctonia solani، آزمایشی بهصورت فاکتوریل در قالب طرح کاملاً تصادفی در سه تکرار انجام شد. تیمارهای این پژوهش شامل سه جدایه P. fluorescens UTPF5 (P5)، P. fluorescens UTPF68 (P68) و P. fluorescens UTPF74 (P74)، قارچ بیمارگر R. solani AG3 و ارقام سیبزمینی شامل آگریا و سانته بود. صفات اندازهگیری شده در این آزمایش شامل وزن خشک ریشه، طول استولون، وزن خشک استولون، وزن خشک غده، وزن تر غده، تعداد غده و تعداد استولون، شدت بیماری در غدههای بذری و فعالیت آنزیمهای GPX و β-1,3-glucanase و محتوای پرولین، مالون دیآلدهید و قند محلول بود. نتایج نشان داد که استفاده از تیمار باکتری توانست در حضور عامل بیماریزا تأثیر معنیداری بر صفات اندازهگیری شده داشته باشد و همچنین شدت بیماری را نسبت به شاهد بهطور معنیداری کاهش دهد. ترکیب تیماری P5 و رقم سانته نسبت به سایر ترکیبات بر صفات اندازهگیری شده تأثیر بیشتری داشت، بهطوری که بیشترین وزن خشک ریشه، طول استولون، وزن خشک استولون، وزن خشک غده، وزن تر غده و تعداد استولون در این تیمار به دست آمد و نسبت به شاهد بهترتیب افزایش 62/0، 86/0، 40/1، 31/2، 37/1 و 63/0 برابری مشاهده شد. استفاده از تیمار P5باعث افزایش فعالیت سیستم آنتیاکسیدانی آنزیم گلوتاتیون پراکسیداز (GPX) و بتا 1 و 3 گلوکاناز (β-1,3-glucanase)، محتوای پرولین و قند محلول به ترتیب به میزان 32/1، 61/0، 2 و 6/2 برابر و از طرف دیگر کاهش پراکسیداسیون لیپید به میزان 57 درصد نسبت به شاهد در شرایط حضور عامل بیماریزا شد.

ارزیابی کارایی Bacillus thuringiensis در مهار زیستی دوبالان قارچ خوراکی در آزمایشگاه و مزرعه
با توجه به اینکه دوبالان پوسیده خوار از آفات داﺋمی سالنهای پرورش قارچ خوراکی هستند، بهناچار بهره بردارن از سموم شیمیایی بهطور مکرر استفاده میکنند. کارگران سالنهای پرورش قارچ و مصرفکنندگان قارچ خوراکی همواره در معرض تهدید سموم شیمیایی هستند، لذا معرفی روشهای غیرشیمیایی و بیولوژیک ضروری است. این پژوهش در قالب دو آزمایش در شرایط آزمایشگاه و مزرعه انجام شد. در آزمایشگاه فرمولاسیون تجاری بایوفلش با ماده مؤثرهBacillus thuringiensis subsp. israelensis با تعدادی از حشرهکشهای رایج روی کمپوست استفاده شد. این آزمایش در آزمایشگاه در قالب طرح کاملاً تصادفی با هفت تیمار شامل دیفلوبنزورون 75/0 و یک در هزار، بایوفلش یک و دو در هزار، اسپینوساد (Tracer®) 3/0 و 6/0 در هزار و شاهد (آب) با 10 تکرار در روزهای مختلف انجام شد. در شرایط مزرعه با توجه به نتایج آزمایشگاه، آزمایش در سه مرحله، محلولپاشی بایوفلش (Bti) یک در هزار روی کمپوست همزمان با تلقیح اسپان، محلول پاشی بایوفلش به میزان 60 میلیلیتر در مترمربع روی کمپوست قبل از خاکدهی و کاربرد بایوفلش بهمیزان 60 میلیلیتر در مترمربع همراه با آب آبیاری شش روز پس از خاکدهی انجام شد. هر سه مرحله در قالب آزمون t، بایوفلش با شاهد (روش معمول) مقایسه شد. نتایج آزمایشگاه نشان داد که همه حشرهکشها میتوانند در کاهش جمعیت دوبالان خسارتزا مؤثر باشند، بهطوری که میانگین کارایی حشرهکشهای دیفلوبنزورون (یک در هزار)، اسپینوساد (6/0 در هزار) و بایوفلش (دو در هزار) روی دوبالان 15 روز پس از تیمار بهترتیب 9/67، 2/74 و 9/79 درصد بود که تفاوت معنیداری باهم نداشتند. نتایج مزرعه نشان داد که کاربرد بایوفلش همراه با آب آبیاری شش روز پس از خاکدهی با 3/79 درصد کارایی، بیشترین تأثیر را دارد.

امکان ذخیره سازی حشرات کامل کفشدوزک Cryptolaemus montrouzieri در دمای پایین
کفشدوزکCryptolaemus montrouzieri مهمترین شکارگر شپشکهای آردآلود در باغات مرکبات و چای شمال ایران میباشد. بهمنظور در اختیار داشتن تعداد کافی از این کفشدوزک و رهاسازی آن در زمانهای مناسب، ذخیرهسازی آن در دماهای پایین ضروری میباشد. در این پژوهش برای بررسی امکان ذخیرهسازی این کفشدوزک، حشرات کامل نر و ماده برای مدت زمانهای 5، 7، 10، 14 و 30 روز درون یخچال (دمای 1±5 درجۀ سلسیوس، رطوبت نسبی 10±60 درصد و تاریکی مطلق) نگهداری شدند و پس از سپری شدن مدت زمان مورد نظر، مرگ و میر آنها شمارش و با شاهد مقایسه شد. سپس، کفشدوزکهای زنده مانده درون ظروف پتری جفت شدند و در داخل یک دستگاه ژرمیناتور (دمای 2±26 درجۀ سلسیوس، رطوبت نسبی 5±60 درصد و دورۀ روشنایی بهتاریکی 16 به هشت ساعت) قرارگرفتند و تا زمان مرگ آخرین فرد، تعداد تخمهای گذاشته شده در هر روز شمارش و یادداشت شد. نتایج نشان داد ذخیرهسازی در دمای پایین زندهمانی، طول عمر و میانگین تخمگذاری روزانه کفشدوزک را بهطور معنیداری کاهش میدهد. با افزایش طول مدت ذخیرهسازی، درصد مرگ و میر حشرات نر و ماده افزایش یافت و بهترتیب از 2/5±39/29 و 8/4±78/25 درصد در تیمار 5 روز به100 درصد در تیمار 30 روز رسید. در همین حال، طول عمر کفشدوزکهای ماده از 2/2±8/99 روز در تیمار شاهد به3/2±5/51 روز در تیمار 14 روز و میانگین تخمگذاری آنها از 06/0± 99/4 تخم/ ماده/ روز در تیمار شاهد به07/0±51/4 تخم/ ماده/ روز در تیمار 14 روز کاهش یافت. بهطور کلی، حشرات کامل این کفشدوزک نسبت بهدمای پایین حساس بودند و ذخیرهسازی بلندمدت آنها (بیش از هفت روز) در دمای پایین (پنج درجۀ سلسیوس) توصیه نمیشود.

کاربرد چند فرمولاسیون زیستی قارچی برای مهار بیماری پوسیدگی سفید سیر در شرایط مزرعه
دو آزمایش مزرعهای برای ارزیابی تأثیر شش فرمولاسیون زیستی جدید در مهار بیماری پوسیدگی سفید سیر ناشی از قارچ Scleotium cepivorum ، در سالهای 1394 و 1395 به اجرا درآمدند. فرمولاسیونهای زیستی بهکارگرفته شده با استفاده از سه گونه قارچ آنتاگونیست (Trichoderma asperellum، T. harzianumوTalaromyces flavus) و یک حامل ارگانیک (سبوس برنج) تهیه شدند. ملاک انتخاب این فرمولاسیونها برای آزمون مزرعهای عملکرد مؤثرشان در مهار پوسیدگی سفید سیر در شرایط گلخانه در مطالعه قبلی بود. آزمایشهای مزرعهای در قالب طرح بلوکهای کامل تصادفی با هشت تیمار (شش فرمولاسیون زیستی، تیمار قارچکش کاربندازیم و تیمار شاهد) در چهار تکرار به اجرا درآمدند. اعمال تیمارها قبل از کاشت و بهروش پوشش بذر (سوخچه) سیر با هر یک از فرمولاسیونهای زیستی، قارچکش یا حامل ارگانیک (برای تیمار شاهد) صورت گرفت. برای اطمینان از وجود آلودگی یکنواخت، علاوه بر اینکه از زمین آلوده به S .cepivorum برای کاشت آزمایش استفاده شد، هنگام کاشت بذور، مقداری مایه بیماری (تکثیر شده روی دانۀ گندم) نیز به بستر کاشت اضافه شد. نود روز پس از کاشت، ارزیابی تیمارها با تعیین میانگین وقوع بیماری و شاخص شدت بیماری در هر تیمار، صورت گرفت. نتایج نشان داد که در سال اول آزمایش اغلب فرمولاسیونهای زیستی مورد آزمون در مقایسه با تیمار شاهد، هر دو فاکتور بیماری را بهطور معنیداری کاهش دادند. فرمولاسیونهای زیستی R.B-T.h-1 وR.B-T.h-2 بهترتیب با داشتن 11/9 درصد و 5/13 درصد بوته بیمار مؤثرترین فرمولاسیونها در مهار پوسیدگی سفید سیر بودند که از لحاظ آماری به اندازۀ تیمار قارچکش بیماری را کاهش دادند. در سال دوم بهدلیل تراکم بالاتر مایه بیماری در خاک، از شش فرمولاسیون زیستی مورد استفاده، چهار فرمولاسیون بهطور مؤثری بیماری را مهار کردند. بهطور کلی نتایج این تحقیق نشان میدهد که این فرمولاسیونهای زیستی جدید میتوانند در مدیریت بیماری پوسیدگی سفید در مزارع سیر، جایگزین قارچکشهای شیمیایی زیانبار شوند.
