جداسازی باکتری‌های دارای خاصیت quorum quenching با قابلیت بیوکنترل Pectobacterium carotovorum subsp. carotovorum

چکیده

تعداد 177 جدایه باکتری کونچر به دو روش استفاده از محیط کشت حداقلی حاوی سیگنال آسیل همو سرین لاکتون از نوع 3-oxo-C6-HSL و محیط کشت‌های عمومی از گیاهان تیره‌ی سولاناسه جداسازی شدند. از بین آنها 11 جدایه متعلق به جنس‌های Pseudomonas، Enterobacter،Acinetobacterو Bacillus فعالیت کونچری بالایی داشتند. به‏ویژه جنس Pseudomonas که دارای بالاترین میزان فعالیت بود. بررسی سازوکار کونچری در این باکتری‌ها نشان داد که آنزیم‌ها و ترکیبات کوچک غیرآنزیمی با فعالیت کونچری در این فرایند نقش دارند. بیشتر این باکتری‌ها فقط دارای فعالیت آنزیمی بودندکه توسط آزمون HPLC تأیید شدند. همچنین برای اولین‌بار جدایه‌ای از باکتری calcoaceticus Acinetobacterکه با روش دوم جداسازی شده بود با توانایی کونچری غیرآنزیمی شناسایی شد. باکتری‌های کونچر عمدتاً باعث کاهش بیوفیلم و کاهش بیماری‌زایی Pectobacterium carotovorum subsp. carotovorumروی سیب‌زمینی و کاکتوس شدند. بنابراین، رابطه‌ی مستقیمی بین افزایش فعالیت کونچری و توانایی بیوکنترلی علیه این بیمارگر وجود داشت، هر چند موارد استثناء نیز مشاهده شد. همچنین، برخی جدایه‌های Pseudomonasبه‌عنوان بهترین عوامل بیوکنترل که براساس فعالیت کونچری علیه بیمارگر مذکور عمل می‌کردند، تشخیص داده شدند.
